At first, we establish the leukemia model induced by γ-ray irradiation in mice and the malignant transformation model in BEAS-2B cell line. With the methods of PCR-single strand conformational polymorphism, real time quantitative PCR, immunoprecipitate and Western blot, gene mutation, mRNA content, the expression and phosphorylation of protein and catalysis activity of SHP-1, SHP-2 were detected in radiation carcinogenesis.
本研究首先建立了γ射线诱发的白血病小鼠动物模型及发生恶性转化的BEAS-2B细胞模型，运用PCR-SSCP、实时荧光定量PCR、免疫沉淀、Western blot等分子生物学及细胞生物学方法，从基因突变，mRNA含量，蛋白质表达量，蛋白质酪氨酸磷酸化水平及酶催化活性等方面多层次地分析了酪氨酸磷酸酶SHP-1、SHP-2在辐射致癌中的作用。
Materials and methods:From June 2008 to November 2008, 45, 249 units whole blood were collected in ACD-B bags in the Blood Center of Zhejiang Province. Then they were centrifugated by 4790×g for 12 minutes to prepare red blood cell suspensions. They were stored at 2-6℃in the refrigeratory. According to the hospital appointments, blood products were irradiated using 25Gy ~(137)Csγ-ray(Gammacell 3000ELAM, MDS Nordion, Canada)for 2 minutes 50 seconds. A sample of RBCs unit, approximately 20ml was collected from each blood bag of 173 RBCS units with a sterile connecting system before and after irradiation to monitor the quality.

以上结果证明随着中药的体内给药治疗，调节和促进了CD4〓T、CD8〓T细胞功能，促进了体内IFN-γ、IL-18的分泌和产生，提示角膜安抗HSK复发的强弱与IFN-γ、IL-18的分泌水平密切相关，结合紫外线诱导后所出现的HSK复发率的结果以及通过FQ-PCR检测的结果可以证实，体内IFN-γ、IL-18分泌生成水平较高的中药18g组、9g组复发率低，其复发后角膜病变混浊度、组织结构损害较对照组减轻。
