In this study, two types of lab-scale reactors, completely-mixed activated sludge and floating-carrier, were set up with feed of ammonium-rich synthetic wastewater. RISA fingerprint, clone library and molecular probe were used to study the relationship between the bacterial community structure variation and the ammonia removal efficiency in both bioreactors. The characteristic species firmly related to autotrophic ammonia removal were decided and the result indicated that the Planctomycete-like organism was the key population fulfilling the deammonification. Several molecular oligonucleofide probes related to deammonification were designed. Based on the probe hybridization results, the microbial mechanism for autotrophic N-removal was revealed, and a hypothesis was raised concerning the unsustainability of activated sludge system. According to the molecular diagnosis results, the operational parameters of the bioreactor were optimized and the ammonia removal efficiency was increased.
